本發明涉及基因工程技術領域,具體地說涉及一類表達于日本血吸蟲表皮的蛋白質、其編碼核苷酸和編碼氨基酸序列以及其作為藥物靶點和疫苗的作用。
背景技術:
目前研究表明,血吸蟲病由血吸蟲的感染引起,在世界上有76個國家,受感染者約1.5億。致病的血吸蟲有3種,既日本血吸蟲(Schistosoma japonicum),曼氏血吸蟲(S.mansoni)和埃及血吸蟲(S.haematobium)。在我國流行的是日本血吸蟲。
研制新的治療血吸蟲病和抗血吸蟲的藥物,首先必須找到合適的藥物作用靶點。血吸蟲的表皮是宿主和血吸蟲相互作用的界面。而組成血吸蟲表皮的蛋白質暴露在這個界面上,對小分子的化學藥物和大分子的抗體藥物而言都是最理想的藥物作用靶點。已經有研究表明治療血吸蟲的化學藥物——比喹酮的作用靶點就是存在于血吸蟲的表皮。另外一些已知的血吸蟲表皮蛋白,例如GST28,paramyosin,已經用于疫苗研究和應用。
發明內容
本發明要解決的技術問題是提供一類表達于日本血吸蟲表皮的蛋白質、其編碼核苷酸、及編碼氨基酸序列日本血吸蟲表皮蛋白,作為可能的藥物作用靶點和候選疫苗方面的作用。
為解決上述技術問題,本發明提供一種DNA序列,它具有序列表中SEQ IDNO.1~SEQ ID NO.23所述序列中任一序列的堿基序列;或者它是在中度嚴緊條件下與序列表中SEQ ID NO.1~SEQ ID NO.23所示的序列中任一條序列雜交的序列;或者它是與序列表中SEQ ID NO.1~SEQ ID NO.23所示的序列中任一條序列有至少70%同源性的序列。
為解決上述技術問題,本發明還提供一種氨基酸序列,它具有序列表中SEQID NO.24~SEQ ID NO.46所述序列中任一序列的氨基酸序列;或者它是與序列表中SEQID NO.24~SEQID NO.46所示的序列中任一條序列有至少70%同源性的序列;或者它是包含有序列表中SEQ ID NO.24~SEQ ID NO.46所示的序列中任一條序列的氨基酸序列。
本發明由于通過大規模的蛋白質組學研究,發現了23個日本血吸蟲表皮蛋白質,這些蛋白質,都是可能的藥物作用靶點和候選疫苗。血吸蟲病的流行給人民身體健康和經濟發展都造成巨大的損失,因此,防治血吸蟲病、開發研制治療血吸蟲病和抗血吸蟲的藥物具有巨大的社會和經濟效益。
圖1是免疫熒光抗體原位雜交實驗結果。
具體實施例方式
下面結合附圖和具體實施例對本發明作進一步詳細的說明。
實施例1日本血吸蟲表皮蛋白的分離和鑒定實驗1.日本血吸蟲表皮的分離純化血吸蟲表皮的純化按照文獻報道的Triton-x-100提取方法進行(參見Oaks,J.A.等人,寄生蟲學雜志,69(3)519-533(1983)。)。表皮蛋白從血吸蟲雄蟲、雌蟲、雌雄混合成蟲和童蟲分離。蟲體用Krebs’-Ringer’s,Tris-馬來酸鹽-緩沖鹽液(KRTM)洗3次,再用KRTM/0.2%Triton X-100溶液懸浮。這個液體和蟲體的混和物置于冰水浴中,在搖床上搖動10分鐘。然后,裝有液體和蟲體的混和物的試管在渦旋混合器上混旋幾次,然后再放回冰水浴中,靜置使蟲體下沉。用吸管吸出上部液體,放置到另一個新管中。這個試管在離心機上用2500×g的離心力,離心15分鐘。得到的沉淀就是血吸蟲的表皮。Krebs’-Ringer’s溶液的配方是0.15M NaCl,6.2mM KCl,1.2mM Na2HPO4,1.2mM MgSO4,10mM Glucose and 20mM Tris-Hepes,pH7.4。Tris maleate緩沖溶液的配方是0.2M Tris and 0.2MMaleicanhyclride,pH7.4。
2.日本血吸蟲表皮蛋白的電泳分離日本血吸蟲表皮用裂解劑裂解,蛋白溶液用SDS-聚丙烯酰胺凝膠電泳分離,條帶切割后再做第二維的在線毛細液相色譜分離以及質譜鑒定。軟件分析得到鑒定蛋白。分離好的血吸蟲表皮,用SDS-聚丙烯酰胺凝膠電泳(SDS-PAGE)緩沖液懸浮,用超聲波促進組織細胞裂解和蛋白溶解,超聲波裂解方法為超聲1秒,暫停3秒,總時間2分鐘。并用100℃加熱5分鐘以使蛋白質變性。然后這些表皮蛋白混合物用SDS-PAGE分離,分離方法按照普通的SDS凝膠電泳方法進行。膠上電泳泳道用刀切割下來,再用刀均等分割成8-12份。接下來做蛋白的膠內酶解。
3.日本血吸蟲表皮蛋白的膠內酶解過程膠內酶解參照文獻(Shevchenko,A.等人,化學年報,68850-858(1996)。)的方法進行。蛋白膠塊用刀切碎,每個小塊大小約為1立方毫米。加入100mMNH4HCO3,洗2次;轉入到試管中;加入10mM二硫蘇糖醇(dithiothreitol,DTT)/100mM NH4HCO3,56度放置反應1小時;冷卻到室溫,離心吸去DTT;加入等體積的55mM碘乙酰胺iodoacetamide/100mM NH4HCO3,室溫/黑暗反應45分鐘;吸去iodoacetamide,加入100mM NH4HCO3,洗2次;b去掉NH4HCO3,加入乙腈縮膠;去掉乙腈,真空抽干;加入100mM NH4HCO3,膠膨脹,去掉NH4HCO3,加入乙腈縮膠;去掉乙腈,抽干;加入含Trypsin的消化緩沖液(50mM NH4HCO3/12.5ng/ul trypsin)冰上放置45min;去掉多余酶液,加入50mM NH4HCO3,以剛覆蓋膠為準;37度,過夜酶切;酶切結束后,從試管中吸出上清液到新管子中,膠中加入60%ACN/0.5%HAC,在水浴中超聲處理15min,再重復一次;吸出上清液到新管子中,在原管子中再加入60%ACN/0.5%HAC,37度40min,吸出上清液,與前面的上清液合并,真空抽氣去ACN。
4.日本血吸蟲表皮蛋白酶解肽段的在線色譜分離和質譜鑒定酶解產生的肽混合物,用真空將溶液體積抽減到30μl。之后,每個組分的肽混合物上樣到一個反相捕獲柱(C18,5μm,300,300μm i.d×5mm,LC Packings),在20μl/min的流速下脫鹽。然后這個捕獲柱和一個分析用的75μm×150mm C18柱(LC Packings)相連接,肽混合物就在200nl/min的流速下,從這個柱子上洗脫進入裝有Protana NanoES的電噴霧離子源的QSTAR pulser i中。Agilent1100毛細液相系統(Agilent Technologies)用來提供流動相A(0.5%乙酸/水)和流動相B(0.5%乙酸/乙腈),線性梯度是60分鐘內5%B到50%B,30分鐘內50%B到90%B,然后是在90%B停留15分鐘。在分析柱后通過一個不銹鋼的兩通(Valco Instrument)連接一個10-μm直徑的PicoTip納噴霧頭(New Objective),加上2500-3000伏的噴霧電壓,以獲得穩定的噴霧。MS/MS串聯質譜譜圖通過信息依賴的采集(IDA)和任務-循環增強來記錄。在每個測定掃描中,選取3-6個信號最強、帶2到3個電荷的離子,用滾動碰撞能量打碎,以獲得串聯質譜的信息。
5.日本血吸蟲表皮蛋白的軟件搜索鑒定用牛胰酶的自裂解片段如([M+H]+)1153.5741,2163.0569和2274.1600作為內標,或者以預先數據庫搜索得到的得分高的肽段的分子量,來校正所有的質譜譜圖。質譜搜索用的數據庫,包括從8420個從EST cluster預測得到的血吸蟲蛋白質序列,以及可能的污染蛋白質序列如牛胰酶的序列、宿主蛋白污染如人蛋白質序列、小鼠蛋白質序列、大鼠蛋白質序列、兔子蛋白質序列和角蛋白序列等。搜索軟件是MASCOT(http://www.matrixscience.com/,MatrixScience)。搜索參數為最大漏切點,3個;monoisotopic;肽電荷數,+1/+2/+3;修飾,半胱氨酸carbamoylmethylation和methionine氧化。搜索結果合并后,用下列標準進行過濾篩選1.對每一個串聯質譜譜圖的搜索結果列表,只提取排位在第一個的結果,其他的則丟棄;2.搜索得到的肽段中如果有KR,KK,RK,RR等序列,則這個肽段丟棄;3.搜索得到的肽段,長度在8個氨基酸以上的保留,小于8個氨基酸的丟棄;4.重復打到的肽段,只計算其中分值最高的,把這些肽段分值加起來,做為蛋白的得分;5.蛋白的得分超過60,就認為是可信的結果。低分的蛋白,手工檢查后保留或者丟棄。匹配到宿主或者牛胰酶、角蛋白的肽段,則丟棄。
實施例2日本血吸蟲表皮蛋白的生物信息學分析和功能注釋
1.同源性和相似性分析日本血吸蟲表皮蛋白序列對GenBank(http://www.ncbi.nlm.nih.gov/)的非冗余數據庫(Non-redundant,NR)做BLASTP比對;日本血吸蟲表皮蛋白序列對Swissprot數據庫做BLASTP同源性比對。根據比對結果得到的與已知其他物種或已知的血吸蟲蛋白的序列相似性,做出日本血吸蟲表皮蛋白的功能注釋。
2.文獻查檢根據已經發表的文獻報道,發現鑒定得到的23個表皮蛋白在蟲體上的位置,有一些的確在血吸蟲表皮上。而其他則是新發現的表皮蛋白。
舉例說明,比如以下表皮分離和質譜鑒定得到的蛋白,根據文獻報道,就是已知的表皮蛋白,而且有些蛋白已經有進行了疫苗和抗原研究。

21.7kDa antigenFK506-binding protein 50(immunophilin)實施例3日本血吸蟲表皮蛋白的實驗驗證為了進一步用實驗的方法驗證用蛋白質組學手段鑒定得到的血吸蟲表皮蛋白,選取了一個Immunophilin(SjFKBP50,GenBank登錄號為AY815389)做免疫熒光抗體原位雜交做蛋白定位分析。
實驗方法血吸蟲成蟲用冷凍包埋劑(OCT)完全包埋,切成5μm厚的切片。用抗Immunophilin的抗體覆蓋切片,在37℃反應60分鐘。該抗體是用重組表達的Immunophilin免疫小鼠制備的單克隆抗體。隨后切片用PBS洗滌,再加入FITC-偶聯的兔抗鼠免疫球蛋白的抗體(1∶40稀釋,用含有0.5mg/ml的Evans藍的PBS稀釋),置濕盒中37℃反應60分鐘。染色結果用Leica DM-RB型熒光顯微鏡觀察并拍照。
圖-所示為免疫熒光抗體原位雜交實驗結果,表皮T;腸道G;下表皮ST。綠色熒光部分是Immunophilin的信號。
該結果證明了Immunophilin確實分布在血吸蟲成蟲的表皮上。這說明前面表皮分離和質譜鑒定的結果是可信的。

2.一種氨基酸序列,其編碼表達于日本血吸蟲表皮的蛋白質,其特征在于它具有序列表中SEQ ID NO.24~SEQ ID NO.46所述序列中任一序列的氨基酸序列;或者它是與序列表中SEQ ID NO.24~SEQ ID NO.46所示的序列中任一條序列有至少70%同源性的序列;或者它是包含有序列表中SEQ IDNO.24~SEQ ID NO.46所示的序列中任一條序列的氨基酸序列。

本發明公開了一類在日本血吸蟲表皮上表達的表皮蛋白,尤其公開了所述表皮蛋白的編碼核苷酸和編碼氨基酸。本發明通過大規模的蛋白質組學研究,發現了一類日本血吸蟲表皮蛋白質,這些蛋白質,都是可能的藥物作用靶點和候選疫苗。血吸蟲病的流行給人民身體健康和經濟發展都造成巨大的損失,因此,防治血吸蟲病、開發研制治療血吸蟲病和抗血吸蟲的藥物具有巨大的社會和經濟效益。
